Biosynthesis of phosphatidic acid from lysophosphatidic acid and palmityl coenzyme A.

چکیده
In the preceding paper evidence was presented for the biosynthesis of lysophosphatidic acid from monoglyceride and adenosine triphosphate (1). Exogenous monoglycende was found to stim.ulate the formation of phosphatidic acid as well as lysophosphatidic acid. This observation suggested that lysophosphatidic acid was being converted to phosphatidic acid and therefore was functioning as an intermediate in the biosynthesis of phosphatidic acid from monoglyceride and adenosine triphosphate. In this paper it is shown that a chemically synthesized lysophosphatidic acid is acylated by S-palmityl coenzyme A to form phosphatidic acid in the presence of a cytoplasmic particulate fraction of guinea pig brain or liver. Lysophosphatidic acid is also degraded to glycerophosphate and orthophosphate by phosphatidases and phosphatases, respectively, in the enzyme preparation.

Partial purification and properties of an acyl coenzyme A:sn-glycerol 3-phosphate acyltransferase from rat liver mitochondria.
The partial purification (6-fold) and properties of a position and substrate specific acyl coenzyme A: sn-glycerol-3-P acyltransferase from rat liver mitochondria are described. The preparation is devoid of acyl-CoA:monoacylglycerol-3-P acyltransferase and lipid phosphomonoesterase activity. All of the glycerol-3-P acylated in the presence of pahnityl-CoA is identified as I-palmityl-sn-glycerol...

متن کاملThe Biosynthesis of Phosphatidic Acid and Lysophosphatidic Acid by Glyceride Phosphokinase Pathways in Escherichia Coli.
Phosphatidic acid has been given a key position in the scheme of animal lipid metabolism because of its ability to serve as a presursor of a variety of animal lipids. The recent reports of the biosynthesis of phosphatidylserine and phosphatidylglycerol (1, 2) in Escherichia coli and other bacteria suggest that phosphatidic acid (in the coenzyme form, cytidine diphosphate diglyceride) will also ...
متن کاملBiosynthesis of phosphatidic acid in lipid particles and endoplasmic reticulum of Saccharomyces cerevisiae.
Lipid particles of the yeast Saccharomyces cerevisiae harbor two enzymes that stepwise acylate glycerol-3-phosphate to phosphatidic acid, a key intermediate in lipid biosynthesis. In lipid particles of the s1c1 disruptant YMN5 (M. M. Nagiec et al., J. Biol. Chem. 268:22156-22163, 1993) acylation stops after the first step, resulting in the accumulation of lysophosphatidic acid. Two-dimensional ...
